IN T RO DU C T IO N Clopidogrel, shown in Figure 1, is a thienopyridine derivative antiplatelet pro-drug used in the prevention of artherosclerotic events. Following oral administration, the dosed compound undergoes hepatic metabolism to give rise to the active thiol-metabolite and the inactive carboxylic acid metabolite. The inactive metabolite accounts for the majority of circulating clopidogrel-related material in humans, while the active metabolite and unchanged pro-drug are present at very low levels. The mechanism of action is derived from the binding of the active thiol metabolite to cell receptor P2Y12, irreversibly inhibiting the platelet activation process.
